PREPARATION OF PLANT-MATERIAL FOR ELEMENTAL ANALYSIS USING ESI AND EELS TECHNIQUES

The optimal preparation of plant material is the prerequisite for elemental analysis using ESI or EELS techniques. Substances must not be extracted or shifted during the preparation procedure. In the present paper some examples are described for preparation techniques permitting elemental analysis. Silica can be analyzed in diatom valves and in Equisetum cell walls after conventional chemical fixation. Water-soluble calcium was precipitated as calcium antimonate or exchanged for insoluble lanthanum. The theoretically best method seems to be cryofixation, followed by freeze substitution or cryosectioning. © 1990.
