UNDERSTANDING AND CONTROLLING TRANSGENE EXPRESSION

Instability of transgene expression is frequently observed in transgenic plants, and is commonly called 'gene silencing', However, silencing that occurs at the transcriptional or post-transcriptional level is not limited to transgenes; endogenous genes can also be suppressed after the transfer of a homologous transgene, Inactivation of transgene expression is often accompanied by an increase in DNA methylation, especially if silencing occurs at the transcriptional level. A comparative study of different silencing systems should help to define the critical requirements for the reliable expression of recombinant constructs in transgenic plants.
