DIRECT AMPLIFICATION OF DNA FROM COLONIES OF BACILLUS-SUBTILIS AND ESCHERICHIA-COLI BY THE POLYMERASE CHAIN-REACTION

摘要
The polymerase chain reaction (PCR) catalyzed by Taq DNA polymerase has been used to amplify genomic and plasmid DNA sequences. In all reported cases, the template has been extensively purified. We show here that genomic and plasmid DNA sequences can be amplified by the PCR directly from single colonies of both Escherichia coli and Bacillus subtilis without any purification of the template. This technique makes the use of the PCR even more applicable in the automation processes, screening and diagnosis. © 1990.
