INCREASED SYNTHESIS OF PROTEINS IS CLOSELY COUPLED TO RELEASE IN THE MYOTROPIC NEUROSECRETORY-CELLS OF RHODNIUS-PROLIXUS

In Rhodnius prolixus, protein synthesis in the 10 neurosecretory cells which produce the myotropic hormone governing ovulation can be measured in in vitro by incorporation of [3H]cysteine. Both the protein content and the rate of protein synthesis of the cells are highest at the time of maximum release. Protein synthesis is stimulated by treatment of the isolated brains in vitro with agents known to provoke release of the myotropin from the cells: ecdysterone and dopamine stimulate protein synthesis in isolated brains from mated ovariectomized females, and the effect of ecdysterone is blocked by phentolamine and phenoxybenzamine, but not by propanolol. The effect of treatment in vitro with ecdysterone is markedly reduced in insects injected earlier with reserpine. Incubation of isolated brains in saline containing 50 mM K+ elevates rates of protein synthesis in brains from both mated and virgin females, an effect which is not diminished by isolating the cell bodies from their terminals in the corpus cardiacum. It is concluded that protein synthesis in these neuroendocrine cells is very closely coupled to events associated with the depolarization of the membrane. © 1990.
