Calf lens MIP [main intrinsic polypeptide] (solvent-extracted from the fiber membranes) and 26,000 band protein (isolated by SDS-PAGE [sodium dodecyl sulfate-polyacrylamide gel electrophoresis]) had very similar amino acid compositions and MW. MIP comprises 2 components immunlogically identrical with the 2 antigens detected in 26,000 band protein. MIP extracts contain 2 antigens in very small amounts which are common to urea-treated fiber and epithelial lens membranes. The 26,000 band protein contains 1 of these antigens. Immunodiffusion, immunofluorescence and SDS-PAGE show MIP is a specific component of the fiber membranes, which is not detected in epithelial membranes. MIP is a marker of differentiation. MIP determinants only occur in the lens demonstrating the organ specificity of the protein.
