ENHANCED ACTIVITY OF THE PLASMA-MEMBRANE LOCALIZED CALLOSE SYNTHASE IN CUCUMBER LEAVES WITH INDUCED RESISTANCE

The resistance of the first true leaf of cucumber to Colletotrichum lagenarium was increased by infecting the cotyledon or the tip of the first leaf with C. lagenarium or tobacco necrosis virus. Microsomes and plasma membranes, prepared from first leaves in which resistance was induced, exhibited increased specific activity of the Ca2+-regulated 1,3-β-glucan (callose) synthase. This enzyme activity is latent in epidermal cells of unchallenged healthy leaves but after activation renders the cells capable of a rapid production of callose-containing papillae. Activation occurs as soon as the Ca2+ permeability of the plasma membrane is perturbed by attempted fungal penetration. © 1990.
