Erythropoietin induces erythrocyte development in vivo as well as some of the biochemical aspects of such development in vitro. When rat bone marrow cells are incubated in the presence of erythropoietin a system involved in the accumulation of extracellular iron in the cell interior is rapidly formed. This induced uptake of iron is inhibited by actinomycin D, puromycin, and cycloheximide, indicating that RNA synthesis and protein synthesis are required for expression of this erythropoietin effect. Part of the protein formed as a result of erythropoietin action was identified as ferritin; the remainder of the system is as yet unidentified, but is known to be neither hemoglobin nor free iron. © 1969.
