SYNERGISTIC KILLING OF EHRLICH ASCITES CARCINOMA CELLS BY ASCORBATE AND 3-AMINO-1,2,4-TRIAZOLE

Ascorbate is shown to be extremely lethal to Ehrlich ascites carcinoma cells in vitro. This lethal effect of ascorbate is synergistically enhanced by 3-amino-l,2,4,-triazole. Catalase, low oxygen tension, and glucose protect the cells from the lethal effects of ascorbate and aminotriazole. Vitamin K3 (as bisulfite = menadione), at a concentration of no more than 1 μg/cm3 (1 ppm), is capable of overcoming such protection. It is concluded that the cytotoxic effect of ascorbate is due to the intracellular production of hydrogen peroxide, and that the enhancement of the ascorbate effect by ATA is due to its inhibition of cellular catalase, thereby inhibiting the ability of the cells to detoxify hydrogen peroxide. © 1969 S.
