STUDIES ON TETRAHYDROISOQUINOLINES (THI) .7. EFFECT OF TRIMETOQUINOL ON CARDIOVASCULAR SYSTEM

The cardiovascular effects of trimetoquinol (AQL-208), the l-isomerof 1-(3,4,5-trimethoxybenzyl)-6, 7-dihydroxy-1,2,3,4-tetrahydroisoquinoline HCI (AQ-110), were investigated and compared with AQ-110 (racemate), AQD (d-isomer and (dl-isoproterenol. The compounds, except for AQD, produced hypotensive, positive inotropic and chronotropic effects as well as peripheral vasodilation. The d-isomer (AQD) of AQ-110 had little effect on the cardiovascular system. The results indicate that the cardiovascular effect of the racemate can be ascribed to the l-isomer. The potency of AQL-208 was about twice that of AQ-110, while it was approximately 1 2.5 that of isoproterenol. In this and preceding papers, it is suggested that the test compounds exert their pharmacological effects mainly by acting as β-sympathomimetics, whereas death caused by toxic doses of the compounds is due to respiratory paralysis. © 1969.
