ALTERATION OF CLOVER PHYLLODY MYCOPLASMA DNA AFTER INVITRO CULTURING OF PHYLLODY-DISEASED CLOVER

Characteristic phyllody symptoms were found to persist in most cases on regenerated plantlets derived by in vitro culturing of phylloid ovaries of clover (Trifolium repens) plants infected with the clover phyllody mycoplasmalike organism (CP-MLO). It was possible to obtain symptom-free plants in a single passage but only at a frequency of 2%. Electron microscopy and DNA hybridization experiments showed the presence of CP-MLO in the diseased regenerates but attempts to transmit the MLO from them to healthy plants by means of vector leafhoppers (Aphrodes bicinctus) were unsuccessful. The loss of vector transmissibility in regenerated plants was associated with either the loss or extensive rearrangement of a 4.8 kb extrachromosomal DNA band present in the CP-MLO-infected plants maintained in the greenhouse through vector transmission. Implications as to the possible role of the extrachromosomal DNA components in pathogenesis are discussed.
