Dicumarol, an uucoupler of mitochondrial oxidative phosphorylation, inhibits the oxidation of succinate by rat liver mitochondria and submitochondrial particles. This inhibition is competitive with respect to succinate and the second power with respect to dicumarol in both systems. The respective inhibitor constants are approximately 4 × 102 μm2 for intact mitochondria (State 3), 10 × 102102μm2 for submitochondrial particles prepared by sonication, and 45 × 102μm2 for succinate-cytochrome c reductase particles. The uncouplers dinitrophenol and the m-chlorophenylhydrazone of carbonyl cyanide also inhibit succinate oxidation in submitochondrial particles in a manner consistant with their inhibition of succinate oxidation in intact mitochondria. It is concluded that all of these uncouplers inhibit succinate oxidation by directly inhibiting the succinate dehydrogenase. © 1969.
