Early changes induced in kangaroo lymphocytes by PHA have been studied by alkaline fast green microspectrophotometry and dry mass measurement. In lymphocytes exposed to PHA, there is a 20-25 per cent decrease in stainability with alkaline fast green, an initial decrease in nuclear dry mass, and an increase in cytoplasmic dry mass. These findings support the hypothesis that PHA induces blastogenesis in lymphocytes through the removal of histone from the DNP complex. © 1968.
