An isotope dilution method has been developed for the determination of hyaluronic acid in tissue samples in the microgram range. The unknown sample is mixed with 14C-labeled hyaluronic acid, proteins are removed by proteolytic digestion and gel chromatography, and the hyaluronic acid is degraded to unsaturated disaccharides by a streptococcal hyaluronate lyase. The disaccharides are then tritiated catalytically and isolated by high-voltage paper electrophoresis. The ratio of 3H to 14C is a linear function of the amount of hyaluronic acid in the sample. The lower limit for the analysis is at present approximately 1 mμeq (0.4 μg) of repeating disaccharide units. The technique has been applied to the analysis of hyaluronic acid in the aqueous humor. © 1969.
