Objective: To test whether Mycobacterium paratuberculosis, the putative etiologic agent of Crohn's disease, was senisitive to clarithromycin. Design: To test the susceptibility of two mycobactin-dependent mycobacteria, M. paratuberculosis and M. avium subsp. silvaticum, to clarithromycin grown in mouse monocyte/macrophage cell line J774A.1. Results: Clarithromycin, 2 and 4 mug/ml, inhibited M. paratuberculosis (80 and 25% of control, respectively) but was less effective against M. avium subsp. silvaticum (80 and 82% of control, respectively) growing intracellularly in the murine macrophage cell line J-774A.1. Conclusions: Clarithromycin shows promise for use in double-blind controlled trials for testing the hypothesis that M. paratuberculosis is etiologically related to Crohn's disease.
