MEMBRANE-BINDING OF CHOLINE-ACETYLTRANSFERASE - UPTAKE AND ACETYLATION OF [CHOLINE-H3 IN RABBIT CORTICAL SLICES

The metabolism was investigated of [3H]-choline accumulated in small slices of rabbit frontal cortex by sodium-dependent uptake. Acetylation (91%) was as efficient as in rat cortical slices (85%), although rabbit choline acetyltransferase (ChAT) lacks the net positive charge thought to localise a critical fraction of rat ChAT at the nerve terminal membrane and to mediate the efficient acetylation of incoming choline. Since ChAT from rat and rabbit brain binds to membrane, after hypo-osmotic lysis of synaptosomes, distribution in vitro seems a better criterion of localization in vivo than net charge. Asymmetric charge distribution or hydrophobicity may explain the disposition of rabbit brain ChAT. © 1979.
