A method is described for evaluating antinociceptive activity in the conscious dog using electrical stimulation of the tooth pulp. Electrodes are implanted under general anesthesia into the dentine of one or other of the upper canine teeth. Tooth-pulp nociceptive thresholds are determined before and at intervals after drug or placebo administration such that each dog acts as its own control. Tests comparing unknown and reference drugs are carried out blind, using cross-over designed assays. The test provides a sensitive, reproducible, and clinically predictive method for evaluating the antinociceptive activities of opioid agonist and partial-agonist/antagonist analgesics and nonopioid centrally acting analgesics. Peripherally acting analgesics are inactive in this test. © 1979.
