CONTROL OF ECDYSTERONE BIOGENESIS IN TISSUE-CULTURES OF TRIANTHEMA-PORTULACASTRUM

Seedling callus of Trianthema portulacastrum was established and screened for phytoecdvsone by a bioassay on the larvae of house-fly Musca domestica. Methods for detection, extraction, separation, and estimation of ecdysterone are described. Effects of various phytohormones and different sucrose levels on growth and ecdysterone production by callus tissues were examined. Naphthaleneacetic acid supported maximum growth of callus and 2, 4-dichlorophenoxyacetic acid (2, 4-D) yielded maximum ecdysterone. Increasing kinetin concentrations were inhibitory to ecdysterone biogenesis. A combination of 2, 4-D with kinetin enhanced growth and ecdysterone production. Gibberellic acid was antagonistic to callus growth enhancedby 2, 4-D and kinetin; however, it stimulated ecdysterone biogenesis. The present study indicates the feasibility for ncreased ecdysterone production in callus cultures as compared to the intact plant. © 1979, American Chemical Society. All rights reserved.
