摘要
The effects of small doses of 1,24,25 trihydroxyvitamin D3 (1,24,25(OH)3D3; 1-10 μg daily by mouth) on intestinal absorption of calcium were measured in 3 normal subjects (5 studies) and 8 patients with disorders of mineral metabolism. No significant change in calcium absorption (assessed on a total body counter by the retention at 7 days of oral 47Ca) was observed after 4 days of treatment in normal subjects or in patients with chronic renal failure. Similarly, 1,24,25(OH)3D3 at 2μg daily for 4 days had no effect on net intestinal calcium absorption assessed by metabolic balance in a patient with idiopathic hypercalciuria. In contrast, an increase in plasma calcium was noted in an anephric hypoparathyroid patient after 40 days of continuous treatment with 1,24,25(OH)3D3 at 10μg daily. These results suggest that 1,24,25(OH)3D3 is much less potent than 1,25-dihydroxycholecalciferol in stimulating the absorption of calcium from the gut and further suggest that the biological effects of 24,25-dihydroxyvitamin D3 in man are not mediated by its conversion to 1,24,25(OH)3D3. © 1979.
