ASSOCIATION OF PHOMA CANKER WITH PHOTOCARCINOGENIC FUROCOUMARINS IN PARSNIP CULTIVARS

Concentrations of naturally-occurring photocarcinogenic furocoumarins, consisting of 5-methoxypsoralen, 8-methoxypsoralen, angelicin, psoralen, and isopimpinellin, were determined in Phoma complanata-infected and healthy foliage, petiole, and root tissue. Samples were obtained from 10 parsnip (Pastinaca sativa) cultivars in field plots at Vineland Research Station in 1987 and from Harris Model in 1989. Reversed-phase HPLC was used with acetonitrile-water as the mobile phase with a concentration gradient. In all tissues examined of all parsnip cultivars, concentrations of total furocoumarins were higher in infected than in corresponding healthy tissues; ranging from 1.2 to 5.0, 1.0 to 5.2, and 21.7 to 1029 times higher in leaves, petioles and roots, respectively. Among cultivars, there was a significant correlation between the ratio of the total furocoumarins in diseased versus healthy leaves and the area under the disease progress curve. High concentrations of furocoumarins also were found in apparently healthy areas 1 cm below diseased root tissue, suggesting that these substances were translocated to healthy parts of the plant.
