摘要
An improved ion-exchange column chromatographic system for the separation and quantitation of neutral mono-, di-, and trisaccharides is described. A two-buffer, stepwise elution system, utilizing a 0.6 × 85 cm column of Dowex 1-X4, eluted with boric acid/2,3-butanediol buffers at a flow rate of 70 ml/cm2/hr and operated at elevated temperatures has been developed for the resolution of complex mixtures of saccharides. Analysis of the column effluents with the aniline/acetic acid/orthophosphoric acid reagent permits quantitation of the saccharides with a precision of approximately ± 5%. © 1969.
