HIGH-FREQUENCY OF POLLEN EMBRYOID FORMATION AND PLANT-REGENERATION IN TRITICUM-AESTIVUM L ON MONOSACCHARIDE CONTAINING MEDIA

The experimental results derived from 15 genotypes of Triticum aestivum L. indicated that pollen embryoid induction frequency in a liquid medium with 0.21 M glucose was 40-750 embryoids per 100 culture danthers and 2-10 times higher than that in the medium with 0.21 M sucrose depending on the genotype used. Fructose or a mixture of glucose, fructose and mannitol also showed improved effects over sucrose. When embryoids were transferred to a filter sterilized double layer plant regeneration medium with 0.21 M glucose and without 2,4-D, 40-90% of the pollen embryoids developed into plantlets; while in sucrose containing medium only 0-30% of the embryoids developed. In one genotype more than 360 pollen plants could be produced from 100 cultured anthers. It seemed that the albino plant frequency was related to the genotypes rather than the medium composition. The factors affecting pollen plant yield are discussed. © 1990.
