CHROMATOGRAPHIC SEPARATION OF PHOSPHONOLIPIDS FROM THEIR PHOSPHOLIPID ANALOGS

Thin-layer Chromatographie separation of phosphono- and phospho-lipid analogs was achieved by using several novel solvent combinations, i.e. mixtures of chloroform with high proportions of 85-92% acetic acid, to which up to 5% methanol may be added when separation of phosphatidylserine from lysophosphatidylethanolamine, phosphatidylinositol and basic phospholipids is desirable. Higher proportions of methanol reduce or eliminate the effectiveness of these solvent systems in separating analogous phosphono- and phospho-lipids. Separation of such analogs of low Chromatographie mobility (e.g. phosphatidylcholine from its phosphono analog) may be further improved, if desirable, by increasing the actual length of the thin-layer Chromatographic run, using simple Chromatographie devices, which were shown here to be applicable in this case. Attempts to separate the phosphatidylethanolamine analogs on silicic acid columns by using this type of solvent systems were partially successful, although promising, and the usefulness of this technique is briefly discussed. © 1969.
