A NEW RAPID ELECTROPHORETIC METHOD FOR DETERMINATION OF DEOXYRIBONUCLEIC ACID BASE COMPOSITION

摘要
A method is presented for the rapid analysis of DNA base composition. After separation from RNA by alkaline hydrolysis a DNA preparation was hydrolyzed with 72% HClO4 for 1 hr. After hydrolysis, HClO4 was removed by evaporation under reduced pressure. Dry powder containing the bases was dissolved in water and separated by agar gel electrophoresis. The spots of each base were cut out with agar gel and after dissolution in warm water were determined spectrophotometrically. The method may be applied to determination of base composition in 50 μg of analyzed DNA. © 1968.
