ISOLATION OF 17ALPHA-ESTRADIOL 17-BETA-D-GLUCOPYRANOSIDE FROM RABBIT URINE, AND ITS SYNTHESIS AND CHARACTERIZATION

Incubation of 17α-estradiol 3-glucuronide with rabbit liver microsomes in the presence of uridine diphosphate glucose, and subsequent removal of the glucuronic acid from C-3 with β-glucuronidase, yielded a monoconjugate which corresponded chromatographically with a radioactive compound found in small amounts in the β-glucuronidase-treated urine of a rabbit injected with 17-β-estradiol-6,7-3H. This material was isolated in crystalline form from the urine of rabbits treated with large doses of estrone benzoate, and was identified as 17α-estradiol 17-β-n-glucopyranoside (Va) by chromatographic and spectroscopic techniques. The structure of this compound was confirmed by chemical synthesis. This is the first demonstration of the occurrence of a steroid glucoside in animals. © 1969, American Chemical Society.
