A total of 66 transgenic plants regenerated from tobacco protoplasts transformed with supercoiled plasmid DNA in different stages of the cell cycle were analysed by Southern blotting. Regenerants obtained from transformation of non-synchronized protoplasts predominantly contain one copy of the introduced foreign DNA in non-rearranged form. The integration pattern of S- or M-phase transformants is quite different, more vector copies are integrated and rearranged forms can be observed in the case of S-phase transformants. In addition, the complexity of the DNA integration pattern of S-phase transformants is influenced by a genomic DNA segment called TBS. © 1990.
