Optimum conditions for various technical procedures in a study of feline viral rhinotracheitis, were investigated. Kitten kidney primary cells and feline lung line cells were equally sensitive for virus assay. Virus was diluted in media supplemented with 10 per cent. calf-serum by 10-fold steps and 4 monolayers per dilution were inoculated. Reading of titrations were carried out after 4 days incubation at 37°C. Virus stocks were harvested when at least 95 per cent. of infected cells showed cytopathic effects. For long term preservation of virus, lyophilization was preferable to freezing and gave reasonable stability for up to 1 year. The virus was quite stable for at least 2 hours at 37°C. plus 18 hours at 4°C. when the suspending medium was buffered with 10 per cent. calf serum. © 1969.
