A modification of the Boyden chamber technic for studying the motility of human polymorphonuclear leukocytes (PMN's) is presented, and various technical factors which might influence the results are discussed. FMN motility was found to vary considerably over the pH spectrum encountered in healthy and diseased tissue. It is possible that tissue variations in pH, through an effect on FMN motility, may play a regulatory role in the development and resolution of certain types of inflammation. Changes in temperature over a fairly wide range had relatively little effect on PMN motility. Ethyl alcohol and caffeine were found to suppress motility at clinically obtainable concentrations.
