2-(4-Phenylpiperidino) cyclohexanol (AH 5183) caused a flaccid paralysis in conscious mice and chicks following oral or parental administration. Experiments using the tibialis anterior muscle preparation of the anaesthetized cat and the isolated phrenic nerve-diaphragm preparation of the rat have shown AH 5183 caused neuromuscular blockade similar in many ways to that caused by (+)-tubocurarine. However, results were obtained to indicate that AH 5183 also impaired transmission in motor nerves by a prejunctional action, probably by inhibiting uptake of choline. Evidence for the latter postulate was obtained using the isolated coaxially stimulated ileum preparation of the guinea-pig in which AH 5183 had a selective prejunctional action on postganglionic parasympathetic nerves. © 1969.
