INFLUENCE OF PROCETOFEN ON LIPID-METABOLISM IN NORMO-CHOLESTEREMIC RATS

Procetofen was administered to rats at a dose level of 10 mg/kg/day. The drug, suspended in gum tragacanth, was given by gavage. For comparison, clofibrate was administered at a dose of 30 mg/kg/day. Control rats were dosed with vehicle. Both substances were hepatomegalic, but procetofen was 10-15% less hepatomegalic than clofibrate. Procetofen administration lowered serum and liver cholesterol levels but raised liver triglyceride levels. Procetofen inhibited the conversion of acetate and mevalonate to cholesterol by suitably fortified rat liver slices; it also inhibited fatty acid synthesis. Cholesterol absorption was similar in the control and procetofen groups and somewhat (17-23%) lower in rats fed clofibrate. The serum plus liver cholesterol pool was of similar size in all three groups of rats. The serum plus liver triglyceride pool was 40-48% smaller in control rats than in the two test groups. © 1979 The Italian Pharmacological Society.
