摘要
Erythropoietin (EPO) plays a key role in O-2 homeostasis by regulating blood O-2-carrying capacity. EPO is produced primarily in the fetal liver and adult kidney. Expression in liver is controlled by DNA sequences immediately surrounding the gene, whereas expression in kidney requires additional sequences located between 6 and 14 kb 5' to the gene. DNA sequences in the 3' flanking region of the gene mediate hypoxia-inducible EPO transcription by binding several nuclear proteins including hypoxia-inducible factor 1, which is induced by hypoaxia in EPO producing and nonproducing cells and activates transcription of other hypoxia-inducible genes.
