SDS-PRESERVATION FOR DEFERRED MEASUREMENT OF EXOPROTEOLYTIC KINETICS IN MARINE SAMPLES

摘要
The effect of sodium dodecyl sulfate (SDS) on exoproteolytic activity was studied in order to test its efficacy as a preservative during deferred analysis of incubated samples. Exoproteolytic activities were assayed using fluorogenic leucine-methylcoumarinyl amide as substrate. Addition of SDS at 1% final concentration entirely inhibited proteolytic activity for both marine pond water and sediment samples. The kinetic parameters, V-m and K-m, of SDS-inhibited samples did not significantly alter during storage at -20 degrees C. The possibility to preserve incubated samples is already proving very useful in field studies where large numbers of exoproteolytic activity measurements cannot be performed immediately.
