Thymidine phosphorylase (thymidine:orthophosphate deoxyribosyltranferase, EC 2.4.2.4) has been approximately 500-fold purified from adult rabbit liver and the kinetics of the conversion of thymine to thymidine, with deoxyribose-i-phosphate as deoxyribosyl donor, are presented. The reaction is subject to inhibition by high concentrations of thymine and this inhibition is modified by urea or low pH. The inhibition persists when the reaction is carried out in 0.5 M NaCl. The effect of both thymine and urea are immediately reversible, and the inhibition by thymine is non-competitive with respect to deoxyribosei-phosphate. © 1968.
