STEPWISE ELONGATION-FACTOR G-PROMOTED ELONGATION OF POLYPEPTIDES ON RIBOSOME WITHOUT GTP CLEAVAGE

摘要
Poly(U)-bound ribosomes [from Escherichia coli] containing presynthesized polypeptide (polyphenylalanyl-tRNA) become competent for binding of the next phenylalanyl-tRNA and subsequent transpeptidation as a result of attachment of elongation factor G [EF-G] with a non-cleavable GTP analog (guanylyl-methylene diphosphonate) and its successive removal from the ribosomes. Hence, EF-G-promoted translocation and resultant elongation of polypeptide can be induced without GTP cleavage. The elongation cycle described can be repeated.
