INACTIVATION OF TRANSFORMING DNA BY ULTRAVIOLET IRRADIATION - A STUDY WITH ULTRAVIOLET-SENSITIVE MUTANTS OF BACILLUS SUBTILIS

The problem of differential inactivation of genetic markers by ultraviolet light was studied with UV-sensitive mutants of Bacillus subtilis. One UV-supersensitive strain (uvss) and three UV-sensitive strains (uvs) proved to be host-cell reactivation negative. When exposed to UV-irradiated DNA, the uvr, but the numbers were considerably larger than those of the uvss strain. A linear relationship, as reported by Bresler et al., was found between the average numbers of hits per genetic unit of length and the UV doses. The number of hits induced by UV in a marker was estimated from the number of tranformants assayed was large in the UV-sensitive marker (asp6+) and small in the UV-resistant markers (met14+ and sfr). This term has been used to interpret the differential inactivation of markers by UV light. © 1969.
