SMALL-VOLUME CUVETTE FOR HIGH-RESOLUTION THERMAL-DENATURATION OF NUCLEIC-ACIDS

A small-volume cuvette and hoider makes it possible to obtain useful high-resolution thermal denaturation profiles on as little as submicrogram quantities of DNA and routine results with approximately 2 μg of DNA. The reduced sample size is especially important in the thermal characterization of DNA restriction fragments. If only the transition midpoint is needed, 0.05 μg of DNA can suffice. With the new microcuvette assembly, multiple samples may be determined simultaneously using a single-beam spectrophotometer equipped with a commercially available cuvette positioner. © 1979.
