摘要
Copy number, the number of expression vectors per host genome, is a key variable in recombinant microbial system study. A quantitative method of monitoring copy number, which is relatively straightforward and quick, is presented here. Cells are first sonicated and then centrifuged to remove debris. The supernatant was filtered to further remove debris and then injected into a Nucleogen 4000 DEAE HPLC system. The ratio of plasmid DNA peak to the chromosomal DNA peak correlated well with the expected variation in the copy numbers of the temperature sensitive recombinant system that was studied.
