OPIATE-RECEPTORS IN SUBSTANTIA NIGRA - ROLE IN THE REGULATION OF STRIATAL TYROSINE-HYDROXYLASE ACTIVITY

Morphine injected into the substantia nigra (SN) of rats failed to change DA metabolism in striatum but blocked the haloperidol induced activation of striatal tyrosine hydroxylase (TH). This blockade was mediated by stimulation of opiate receptors in SN because it was antagonized by naltrexone. The binding of 3H-enkephalinamide to opiate receptors in SN was not altered after lesion of the nigrostriatal DA pathways with 6-hydroxydopamine but was decreased (approximately 70%) after brain hemitransection. These results suggest that intranigral morphine prevents the haloperidol induced activation of striatal TH by modulating the release of endogenous transmitters or modulators on DA neurons. © 1979.
