MOLECULAR RELATIONSHIPS OF THE MITOCHONDRIAL-DNA OF OPHIOSTOMA-ULMI AND THE NAN AND EAN RACES OF O-NOVO-ULMI DETERMINED BY RESTRICTION-FRAGMENT-LENGTH-POLYMORPHISMS

Mitochondrial (mt) DNAs from 11 isolates of Ophiostoma ulmi, 12 isolates of O. novo-ulmi EAN race and 12 isolates of O. novo-ulmi NAN race, obtained from 20 different countries, were compared by analysis of fragments produced by digestion with restriction endonucleases Bcl I, EcoR I and Pou II. The size ranges of the mtDNAs of the three subgroups were generally distinct i.e. O. ulmi (74-78 kb), O. novo-ulmi EAN race (65-69 kb) and most isolates of O. novo-ulmi NAN race (50-59 kb). A total of 21 mtDNA types was distinguished, 10 in O. ulmi, 4 in O. novo-ulmi EAN race and 7 in O. novo-ulmi NAN race. Dendrograms produced from restriction endonuclease profiles demonstrated that generally the mtDNAs of isolates within a subgroup (O. ulmi, EAN or NAN) were more closely related to each other than to mtDNAs of other subgroups and that the mtDNAs of the EAN and NAN races of O. novo-ulmi were more closely related to each other than to the mtDNAs of O. ulmi. MtDNAs of two NAN isolates with typical NAN nuclear profiles had sizes (ca. 72 kb) dose to the O. ulmi mtDNA size range and, in the dendrograms, clustered with the O. ulmi isolates, suggesting possible rare introgression of O. ulmi mtDNA into the O. novo-ulmi NAN race.
