ENZYME-SUBSTRATE ASSAY FOR THE QUALITATIVE DETECTION OF MICROORGANISMS BY ION MOBILITY SPECTROMETRY

A relatively new concept is explored where the potential for ion mobility spectrometry (IMS) is investigated for the detection and determination of living microorganisms. A hand-held IMS instrument embodies the analytical concept and is an atmospheric pressure vapor detector. Neutral vapor analytes are ionized by a Ni-63 ring and the ions' time of flight in an atmospheric pressure drift tube are recorded. Advantage is taken of the inherent enzymes found in microorganisms and an exogenous tailored substrate was provided to initiate the desired biochemical reaction. The substrate was ortho-nitrophenyl-beta-D-galactopyranoside, and the product, ortho-nitrophenol (ONP), has a considerable vapor pressure. ONP can be detected in the negative ion mode of the IMS which indirectly signals the presence of bacteria. Detection limits of almost-equal-to 10(4) Escherichia coli bacterial cells in 5 min and 3300 E. coli cells in 15 min were realized. The results suggest a new application of IMS in the screening of bacterial contamination in community water and wastewater testing situations.
