BIOCHEMICAL STUDIES OF METHOXYINDOLES . DETERMINATION OF N-ACETYL-5-METHOXYTRYPTAMINE (MELATONIN) BY SPECTROFLUOROMETRY

Among the indolic compounds with a methoxy group in the 5 position, the N-acetyl-5-methoxytryptamine (melatonin) presents particular interest. It is synthetised in the pineal gland, and its urinary excretion has physiological and pathological variations that have not yet been sufficiently clarified. A sensitive and specific method for the spectrofluorometric determination of melatonin is presented, based on the analytical study of indolic derivatives: fluorescence spectra and its variation with pH, solvent extraction in the function of the pH, chromatography. The successive steps of the technique are the following:. 1. - First extraction with chloroform at pH 10. 2. - Evaporation of CHCL3 and dissolution of the residue with 0.1 N HC1. 3. - Second extraction with chloroform at pH 7.4. 4. - Second evaporation of chcl3 and dissolution of the residue with 0.1 N HC1. 5. - Purification on alumina, at pH 10. 6. - Spectrofluorometric reading. The technique may be applied to urine or tissues (e.g., pineal gland). Our first results agree well with those found by various authors. We shall shortly report on a spectrofluorometric technique method for the determination of 5-methoxytryptamine along the same principles. © 1969.
