SURVIVAL OF PSEUDOMONAS SPP IN AIR-DRIED MINERAL POWDERS

Limited evidence suggests that some minerals enhance the ability of bacteria to survive desiccation. Our objective was to use a wide variety of well-characterized minerals to determine what effect mineral type has on the survival of Pseudomonas spp in air-dried powders. Two strains of Pseudomonas fluorescens (2-79RN(10), and W4F393) and a fluorescent Pseudomonas sp. strain Sg-l were suspended in phosphate buffer or 10% lactose and mixed into Ca2+-saturated kaolinite, montmorillonite, vermiculite, zeolite, talc or pyrophyllite. Preparations were air-dried and stored at 20 degrees C and 15% relative humidity. When lactose was absent from the preparation, minerals with a smaller average particle size (i.e., montmorillonite, zeolite and vermiculite) enhanced bacterial survival more than minerals with a larger particle size (i.e., pyrophyllite, talc and kaolinite). The survival of three Pseudomonas spp under desiccation stress was, therefore, positively correlated with mineral surface area regardless of lattice type. The addition of lactose enhanced the survival of both 2-79RN(10) and S8-1 when incorporated with pyrophyllite, tare or kaolinite, but did not increase survival when Formulated in high surface area minerals. The use of minerals with a smaller average particle size or the addition of lactose to minerals with larger size particles should be considered when attempting to promote bacterial survival in air-dried mineral powders.
