Glucose oxidase, in its catalysis of the reaction between glucose and oxygen, is shown to be consistent with the reactions (1) E + G →- RL; (2) RL → R + L; (5) RL + G →R + L + G; (3) R + O2 → E.H2O2; and (4) E. H2;O2; →-ʹE + H2O2, where E is oxidized enzyme, G is β-D-glucose, R is reduced enzyme, and L is glucono-lactone; RL and E H2;O2; are complexes involving enzyme and products. The rate constants k-. and are determined at 0 and 15° At 30°, k2; and k5; become too large to determine and the three-parameter mechanism involving only k1;, and k4; reproduces the data. Oxygen-uptake data and steady-state stopped-flow experiments lead to identical results, within experimental error. © 1969, American Chemical Society. All rights reserved.
