DOUBLE-REDUCTION OF A(1) ABOLISHES THE EPR SIGNAL ATTRIBUTED TO A(1-) - EVIDENCE FOR C2 SYMMETRY IN THE PHOTOSYSTEM-I REACTION-CENTER

Illumination of Photosystem I preparations at room temperature at pH 10.0 in the presence of dithionite has been shown to double-reduce the bound phylloquinone electron acceptor A1 (Setif, P. and Bottin, H. (1989) Biochemistry 28, 2689-2697). The asymmetric EPR signal (g = 2.0048, DELTAH ptp = 0.95 mT) photo-accumulated by illumination of Photosystem I at 205 K is removed by such treatment, confirming the attribution of this EPR signal to A1-. Photoaccumulation of A0- and A1- EPR signals at 230 K in Photosystem I frozen at pH 10.0 approaches a maximum of 4 spins per P700+, suggesting that Photosystem I exhibits the C2 symmetry of other types of photosynthetic reaction centre.
