PREPARATION AND PURIFICATION OF BETA-AMYLOID(1-43) VIA SOLUBLE, AMIDE BACKBONE PROTECTED INTERMEDIATES

摘要
We report a new synthetic strategy for the preparation of the amyloidogenic protein fragment beta A4, through an easily purified (and characterized) intermediate of high aqueous solubility. This novel intermediate consists of beta A4(1-43) with backbone amide protection, provided by the N-(2-hydroxy-4-methoxybenzyl) group, on residues Phe(20) and Gly(25,29,33,38). The new strategy enabled derivatized beta A4(1-43) to be isolated and purified by standard reverse-phase HPLC procedures and characterized by electrospray mass spectrometry (ESMS).
