TURBIDIMETRIC MEASUREMENT OF CELL BIOMASS OF PLANT-CELL SUSPENSIONS

摘要
A new method using a specialized cuvette in a spectrophotometer was developed for turbidimetric measurement of cell concentration in plant cell suspensions. The spectrophotometer was able to mix plant cell suspensions in the cuvette almost homogeneously, giving a stable optical density (O.D.). When mixing was done by agitation with a magnetic stirring bar in the cuvette, the O.D. decreased at slow agitation speeds as the cell aggregates sedimented. At high agitation speeds, however, the O.D. increased as cell aggregates were broken. At an optimal agitation speed (2,500 rpm) neither aggregate sedimentation nor mechanical damage occurred and the O.D. was consistent and reproducible. In cell suspensions of Catharanthus roseus and Oryza sativa, close relationships were obtained between the values of O.D. and dry cell weight.
