INSULINOGENIC MEDIATION OF SCHEDULE-INDUCED POLYDIPSIA

It has recently been suggested that schedule induced polydipsia (SIP) might be due to hypoglycemia mediated via neurogenic increases in insulin secretion. This hypothesis was tested in rats made polydipsic on a fixed time 60 sec schedule. Surgical (coeliac ganglionectomy) and chemical (propranalol) blockade of β-adrenergic pancreatic influences had no effect on SIP. Peripheral parasympathetic blockade with methyl atropine did decrease SIP in a dose-related fashion, but since various selective vagotomies failed to reduce SIP, the atropine effect was probably non-specific. In addition, exogenous insulin injections failed to potentiate SIP, and blood measures taken before and during SIP sessions failed to reveal the hypoglycemia predicted by the insulin hypothesis. We conclude that it is highly unlikely that SIP is mediated through autonomic influences on insulin secretion. © 1979.
