摘要
A simple method is described for visualizing submicrogram amounts of the CNBr cleavage products of proteins on sodium dodecyl sulfate-polyacrylamide gels. The peptide fragment mixture is conjugated with the fluorogenic reagent 2-methoxy-2,4-diphenyl-3-(2H)-furanone prior to electrophoresis. The fluorescnet peptide bands are visible under UV light, avoiding the need for fixation and staining. The determination of the structural homology of 2 immunologically related proteins [rat skin prolyl hydroxylase and a cross reacting protein] is presented to illustrate this methodology.
