他汀类药物对参与动脉粥样硬化的主要细胞成分的直接作用

摘要
他汀类药物是降脂药,它的作用机理是抑制3-羟基-3-甲基戊二酰辅酶A(HMG-CoA)还原酶,该酶催化HMG-CoA转化为甲羟戊酸。甲羟戊酸的代谢产物是真核细胞一系列生物活动的关键,他汀类抑制甲羟戊酸代谢通路后可产生多种效应。已发现他汀类可抑制血管平滑肌细胞(VSMC)和巨噬细胞迁移、增殖;减少VSMC中白细胞介素-6和诱导型一氧化氮合酶表达;改善内皮功能;上调内皮型一氧化氮合酶表达。他汀类的上述作用与血浆胆固醇水平无关,可以被内源性甲羟戊酸和某些类异戊二烯物质所阻断。因此除降脂作用外,他汀类药物还可作用于参与动脉粥样硬化的主要细胞成分。发挥直接的抗动脉粥样硬化作用。
