新型PD模型研究进展

摘要
帕金森病(Parkinson disease,PD)是老年人第二大常见的神经系统退行性疾病,仅次于阿尔茨海默病,严重影响患者的生活质量。尽管对PD的发病过程有很多研究,但其发病机制尚未完全清楚。近20年来,动物模型的使用使PD研究取得了突破性的进展,这为更加深入了解PD的发病过程、病因病理和分子机制提供了很大帮助。最早建立的PD模型是利用利血平,随后逐渐出现了甲基苯丙胺PD模型、6-羟基多巴胺(6-hydroxydopa,6-OHDA)PD模型、1-甲基-4-苯基-1,2,3,6-四氢吡啶(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPTP)PD模型、鱼藤酮PD模型等,其中以6-OHDA PD模型和MTPT PD模型应用较多,而鱼藤酮作为一种新型的诱导药物且所造模型产生路易小体(Lewy bodies,LB)受到广泛关注。以基因学为基础创建的PD动物模型在揭示引起多巴胺(dopamine,DA)能神经元死亡的分子机制和发现潜在的治疗靶点方面具有重要意义。本文就近年来出现的以鱼藤酮为诱导药物和以基因学为基础建立的PD模型的相关研究进展进行综述。
