人外周血淋巴细胞化学发光测定的方法学研究

摘要
人外周血淋巴细胞(PBLC)受ConA或PHA刺激后, 能产生短暂的、Luminol依赖性的化学发光(CL)反应。实验表明, 在有限稀释范围内, 淋巴细胞、ConA或PHA浓度越高, 淋巴细胞化学发光(Ly-CL)的强度越高, Ly-CL强度依赖于淋巴细胞、ConA或PHA浓度。Luminol在1×10-4M时, Ly-CL最强。5种细胞稀释液中, 以pH7.2~7.4、含1~3%小牛血清(CS)的HBSS用于Ly-CL测定最佳。新分离的淋巴细胞放置4℃ 22小时、22℃ 8小时或37℃ 2小时内, Ly-CL变化不显著。Ly-CL测定具有简便、快速、灵敏度高、重复性好等优点, 有可能成为研究淋巴细胞早期激活或功能测定的一种新手段。
